ENA is a combination of different influences, that one included. The design is also draws heavily from Mariya Takeuchi, Peruvian school uniforms, and 90s PC aesthetic (see old Finder icon). Everything is influenced by things that came before and experiences of the artist.
Twitter doesn't count as canon
If it's not in the finished product, it's not cannon. Same reason that Dumbledore isn't gay and Bridget isn't trans. Creators had 20 years to put that stuff into print / the game, and only released it as a inconsequential footnote to appease a baying mob on twitter.
ENA has never shown romantic or sexual desire despite numerous interactions. Her closest relationship is as a doormat for moonie in a platonic way. ENA has all the sexuality of SpongeBob SquarePants.

Joel didn't need to put ENA's "sexuality" in the viewers' faces, he's shown ENA's disinterest in romance/sex by the character's actions. ENA loves turron, and only turron. Until Joel writes otherwise, that's canon.
