Is it possible that Stan and/or Ditko envisioned them as Peter's great aunt and uncle? And then somehow along the way that got muddled and the more direct relation was established more firmly by someone
Let's say Pete's parents were 40 something when he was born. Aunt May would be like, in her 60s when we first see her. What do you think a 60 year old would look like in the 60s? She could have been born in the past century!
If Marvel kept killing off all the wasted supporting cast, the characters would just be in costume 24/7.

omd had two route, either kill aunt may, the last thing that held onto peter's past being the maternal character he needs to outgrow.
Or remove mary jane and accept he will never move on, mature, be financially indpedent adult who has to deal with the maturity of life and developing a family.
She should have stayed dead 30 years ago. It just feels cruel to keep her alive while Ben is one.
