It very well could be the actual sincere reason, but given all we've seen in leaks and episode descriptions and clips, there's nothing story-wise that would match up with them being changed to twins instead of just friends, not even the theme that said friendship. Anons who think they changed it because the Gay of Thrones writer wants a gay couple or because execs think twins sell after Gravity Falls or even for multiple reasons have some credit to them, but really we won't know exactly why until the show actually airs
In the original show, Buster X Plucky don't have the Bugs X Daffy dynamic. For one thing, Daffy is laser focused on Bugs as the Looney Tunes star who stoles his spotlight, since Daffy was the headliner first. Plucky does sometimes resent both Babs AND Buster, but he's not as fixated on it, his spotlight wasn't stolen, and some of that is just directed at everyone who isn't him.
I think Plucky X Hampton makes more sense as a fujo ship in the original show. There's more focus on that.
Original show also had the bunnies with no relation and dating, this show just has a lot of differences

This post and a bunch of old Animaniscs vs. Animaniacs 2020 reboot posts here on Cinemaphile are some strange a desperate revisionism to just how incredibly fricking different these two reboots and their characters (just "so far" in the case of Looniversity, but still) are turning out to be from the original shows and characters.
It is okay for reboots to be different from the originals. You don't have to lie and pretend the originals were totally different and viewed totally differently to put them in line with the reboots for some reason. Maybe it's just because nobody has watched Tiny Toons or Animaniacs from three decades ago when they were four years old.

Yes you could call Babs and Buster more on again off again and casual which for kids their age made sense, but they definitely kissed, dated, mutually crushed pretty often.
