None of the examples posted ITT are equivalent.
None of these are things that people will know about even if they know nothing about the comics or cartoons said spoilers come from.
Yes, because it's been referenced everywhere.
I knew about it long before I ever watched the original trilogy because it's been referenced everywhere for fricking decades.
This is probably THE most referenced spoiler in modern media.

There are videos posted to this day of people who have never watched star wars being surprised by the reveal
Yea, because as we all know, nothing is more legit than react videos.
I could kinda see it. Vader spoofs aren't as popular/prominent as they once were.
They were everywhere in the 90s, there were a few in the 00s (I can only think of Darth vegan though), and I doubt anyone cared in the 10's
If you mean just the scene itself, sure, like I could see zoomers or whoever going nuts for the Thanos Snap despite it already being a known quanitity just because it's a dramatic scene but nobody is shocked by the reveal itself. Most Zoomers probably know Vader more as Anakin than the opposite at this point, it's not a reveal, every appearance with him the past couple years is built entirely around the fact that you KNOW his relation to the Jedi and his children.
The most famous spoiler in comic book history is Captain Atom is Monarch.
The tldr of it is the 1991 DC event Armageddon 2001 was built around the mystery of which hero in 10 years will become the super villain Monarch and kill all the superheros.
The twist somehow leaked and then there was a run of speculators buying up back issues of Captain Atom forcing DC to panic and change the twist ending.
DC changed Monarch to Hawk of Hawk and Dove even tho their tie in showed that Hawk couldn't be Monarch.
So the book is rewritten and Monarch kills Dove, Hawk kills Monarch and finds out he is Monarch, puts on Monarch's costume to become Monarch and then tries to kill all the heroes but is thwarted.

...And people know that Darth Vader is Luke Skywalker's father even if they don't know a single thing about Star Wars?

That's right, no amount of cope will ever mean that old out of touch granny akshully knew about the twist the whole time
Old out of touch grannies wouldn't know about any twists unless it's from something from before the 1970s anyways.

Bro, pretty much every adult animated show had a Vader spoof in the 00s and early 10s.

Given how few people read comics compared to watching movies there aren't many among normal people.
Among comics fans it's basically the secret identities of heroes/villains as there are almost no mysteries.
Alucard is Dracula backwards
